Determination of anti-cancer constituents in oplopanax horridus and oplopanax elatus

Abstract

A rapid and reliable RP-HPLC method for the analysis of 3αhydroxylup -20(29)-ene-23,28-dioic acid(1) and 3αhydroxylup-20(29) -ene-23, 28-dioic acid-3-O-βD-glucoside (2) in leaves of Oplopanax horridus and O. elatus was established, and their contents changes between species and different cultivated places were compared. The established analysis method presented good results and could be used as a method for the quality control of leaves of O. horridus and O. elatus. Meanwhile, the inhibitory effect of 1 and 2 on human hepatoma carcinoma cell (HepG-2) were examined and their IC 50 were determined to be 41.15 μM and 120.06 μM indicating that the anti-cancer activity of 1 was stronger than that of its glycoside. Moreover, the inhibitions of 1 on human colon cancer cell line (HCT116), human lung carcinoma cell line (NCI-H460) and human gastric cancer cell line (MGC803) were further tested, and the IC50 were determined to be 21.40 μM, 22.80 μM and 21.26 μM, respectively. While the inhibition of 1 on human pancreatic cancer cell line (PANC-1) was insignificant, indicating 1 possessed selectivity for the anti-cancer activity.
